Structural Characterization

HCG is a heterodimeric glycoprotein composed of two non-covalently associated subunits: a 92-amino acid α-subunit (common to the glycoprotein hormone family) and a 145-amino acid β-subunit. The α-subunit contains five disulfide bonds and two N-linked glycosylation sites (Asn⁵² and Asn⁷⁸). The β-subunit contains six disulfide bonds, two N-linked glycosylation sites (Asn¹³ and Asn³⁰), and four O-linked glycosylation sites in the C-terminal extension (Ser¹²¹, Ser¹²⁷, Ser¹³², Ser¹³⁸).

  • Subunit Architecture: α (92 aa) + β (145 aa), non-covalent heterodimer
  • Disulfide Bonds: 5 (α-subunit) + 6 (β-subunit), cystine knot motif
  • Glycosylation: 4 N-linked + 4 O-linked sites; sialic acid-capped
  • Molecular Formula: Glycoprotein — variable with post-translational modifications

Physicochemical Properties

  • Appearance: White to off-white lyophilized powder
  • Isoelectric Point: pI ~3.5 (acidic, due to sialic acid residues)
  • UV Absorption: λmax 280 nm (Trp/Tyr residues)
  • Solubility: Freely soluble in aqueous buffers
  • Thermal Stability: Store lyophilized at 2–8°C; avoid repeated freeze-thaw cycles in solution
  • SDS-PAGE: α-subunit ~22 kDa, β-subunit ~38 kDa under reducing conditions (apparent MW includes glycan mass)
